Bio-Synthesis offers Photocleavable PC Spacer Oligonucleotide Modification
at various scales. Photocleavable PC Spacer has a non-nucleosidic moiety that can
be used to couple to the terminal end of an oligonucleotide through a UV photocleavable
C3 spacer, as well as insert such a spacer internally. An example is the use of
PC Spacer to incorporate a photocleavable 6-FAM tag onto the 5’-end of oligonucleotides
immobilized on glass slides. These fluorescently-labeled oligo arrays are then
UV irradiated in order to test the efficacy of photocleavage in removing the 6-FAM
tag from these oligos, as part of developing sequencing-by-synthesis applications.1
Another possible use of this photocleavable spacer is to 'cage' oligonucleotides.
Caging allows the biological activity of a species to be suppressed until released
by an external agent - in this case a flash of UV light. This would give researchers
exquisite spatial and temporal control of the concentration of the active species.
Photocleavable spacer modified oligonucleotides could be used to tether an antisense
oligo to a dextran, which is too large to pass through the nuclear pores. Photocleavage
would then be used to release the oligo, thereby decoupling antisense effects within
the cytoplasm from antisense effects within the nucleus.

Purification:
Spacer modified oligonucleotide can be accomplished by various methods. Desalting
or cartridge purification is acceptable, however, additional purification by HPLC
is strongly recommended.

Cleavage Protocol
Cleavage occurs by irradiation with near-UV light (300-350 nm). Try using a UV lamp
at a distance of 15 cm (emission peak 365 nm, 300 nm cut-off, 1.1 mW intensity at~31
cm).
