Dual Biotin Oligonucleotide Modification

Dual biotin can be incorporate at 5' or 3' end of an oligonucleotide result in two functional biotin groups which act to increase binding affinity of biotin-streptavidin. This is especially useful when immobilize the biotin-labelled DNA/RNA target sequence on the beads and incubate with the cell extract. Dual biotin labelled oligo keep biotinylated DNA/primers on the beads during heating after higher temperatures. Study have shown dual biotin prevents or effectively reduces leakage of biotinylated DNA from beads during heating1. Oligonucleotides are stable in solution at 4°C for up to 2 weeks. Properly reconstituted material stored at -20°C should be stable for at least 6 months. Dried DNA (when kept at -20°C) in a nuclease-free environment should be stable for years.
