BlackBerry® Quencher - BBQ-650® Quencher Oligonucleotide Labeling
Bio-Synthesis offers BBQ650 quencher oligonucleotide modification. BBQ650 exhibits a broad absorption profile from 550nm to 750nm, centered at 650nm. BlackBerry® Quencher 650 (BBQ-650®) is ideal for the quenching of long-wavelength reporter fluorophores. It is an excellent FRET quenching probe beause has long spectral overlap with long-weavelength fluorophores such as Texas Red, ROX, Cy3, Cy5 and Cy5.5 thus allowign efficient FRET quencing. Especially Cy5.5 was quenched with 99% efficiency when separated from BBQ-650 by ten base pairs (ca. 40 Å) in a non-contact hybrid probe.
BBQ650 quencher also an excellent contact-mode quencher such as molecular beacon. It offer effective quenching of some of our popular long wavelength dyes like Texas Red, FAM, Redmond Red, Cy3, Cy5, Cy5.5 and DyLight dyes. BlackBerry® Quencher 650 (BBQ-650®) exhibit high affinity for Cy dyes. Signal-to-background ratios of >90 and >88 for Cy5 and Cy5.5, respectively, were observed in molecular beacons.
BBQ650 is chemically resistant to both oligonucleotide synthesis reagents such as iodine, TCA or deblocking solutions such as ammonia, AMA. As result, BBQ650 is an ideal quencher for longer oligos (>50 bases) than BHQ-2 or BHQ-3, as the latter are chemically less stable and degrade with exposed to oligo synthesis and deprotection condition for long periods of time.
We offer BBQ-650 products for the 3' and 5' termini, as well as BBQ-650-dT for inclusion within the oligonucleotide sequence, with the following properties:
- Quenches the fluorescence of long wavelength dyes (Cy3, Cy5, Cy5.5, Texas Red and FAM)
- Excellent as FRET-mode quencher or contact-mode quencher such as molecular beacon
- Absorbance maximum at ~650nm
- Quenching range - 550-750nm
- Compatible with standard oligo synthesis chemistry
- Compatible with regular deprotection but requires mild deprotection with AMA at room temperature
- Available for 3', 5', and internal substitution
- More stable than BHQ-3

Oligonucleotides are stable in solution at 4°C for up to 2 weeks. Properly reconstituted material stored at -20°C should be stable for at least 6 months. Dried DNA (when kept at -20°C) in a nuclease-free environment should be stable for years.
