Acrydite Oligonucleotide Modification
Acrydite can be place placed at the 5' end of an oligonucleotide. The acrylamide group of acrydite modified oligo is covalently react with thiol-modified surface or can be incorporated into polyacrylamide gels during polymerization via a thioether bond. The immobilization leaves the oligonucleotides available for hybridization. The Acrydite modification is insensitive to heat and other denaturing agents. Acrydite-modified oligos are stable in aqueous solutions over a wide range of temperature and pH. Oligonucleotides are stable in solution at 4°C for up to 2 weeks. Properly reconstituted material stored at -20°C should be stable for at least 6 months. Dried DNA (when kept at -20°C) in a nuclease-free environment should be stable for years.
