6-Thio-Guanosine (6-thio-G) is an photoreactive RNA base has been often use in ribozyme and siRNA research , as well as RNA-protein interactions. In one of the study has shown RNA can isolated by treatment of cell s in the culture with 6-thiogunosine or 4-thiouridine followed by affinity chromatography on mercurated cellulose columns. 4-Thiouridine (4-thio U) and 6-thioguanosine (6-thio G) are specifically used for identification of components of RNA-binding by its ability to site-direct cross link to the center of ribosome. This photo-reactive base can form the so-called zero-length cross-linking. A sigle 6-thio G base in an RNA molecule does not change its spatial structure when forming RNA-RNA cross-link. this modification allows one to idetnify direct contacts between nucleotides of rRNA and RNA ligands.
